ACTOne PDE Stable Cell Lines

eEnzyme's ACTOne™ PDE Stable Cell Lines are live-cell functional assay systems designed for quantitative analysis of phosphodiesterase (PDE) activity and inhibitor screening. Based on the proprietary ACTOne™ membrane potential biosensor platform, these engineered cell lines combine recombinant human PDE expression with a modified cyclic nucleotide-gated (CNG) fluorescent reporter, enabling rapid and sensitive measurement of intracellular cAMP signaling in living cells.

Phosphodiesterases (PDEs) are a family of enzymes that hydrolyze the second messengers cyclic AMP (cAMP) and cyclic GMP (cGMP), thereby regulating the intensity and duration of numerous cellular signaling pathways. The mammalian PDE superfamily consists of 11 families (PDE1–PDE11) with distinct substrate specificities, tissue distributions, and biological functions. Because PDEs control signaling involved in inflammation, neurotransmission, cardiovascular function, metabolism, and smooth muscle contraction, they have become important therapeutic targets for diseases such as COPD, asthma, heart failure, pulmonary hypertension, Parkinson's disease, Alzheimer's disease, schizophrenia, erectile dysfunction, and cancer.

Unlike traditional biochemical enzyme assays using purified proteins, ACTOne™ PDE Stable Cell Lines measure PDE activity directly in living cells while preserving native intracellular signaling pathways. Inhibition of PDE activity increases intracellular cAMP levels, producing a fluorescent response that can be quantified using standard fluorescence plate readers. This homogeneous, no-wash assay format offers excellent sensitivity, broad dynamic range, and high reproducibility, making it well suited for both routine pharmacological studies and high-throughput screening.

Biological Significance of Phosphodiesterases

Phosphodiesterases are essential regulators of intracellular signaling because they terminate cAMP and cGMP signaling by catalyzing their hydrolysis. Different PDE isoforms control distinct physiological processes, including cardiac contractility, neuronal communication, immune responses, metabolism, smooth muscle relaxation, and hormone signaling. Selective inhibition of specific PDE family members has already produced successful therapies, such as PDE5 inhibitors for erectile dysfunction and pulmonary arterial hypertension and PDE4 inhibitors for inflammatory airway diseases. Additional PDE families continue to be actively investigated as therapeutic targets for neurological disorders, metabolic diseases, fibrosis, and cancer.
