About Human Angiotensin II Receptor Type 1 (AT1R) Stable Cell Line

Angiotensin II Receptor Type 1 (AT1R), also known as AGTR1, is a Class A G protein-coupled receptor (GPCR) that serves as the primary receptor for angiotensin II, the principal effector peptide of the renin-angiotensin system (RAS). Upon activation by angiotensin II, AT1R primarily couples to Gq proteins, leading to phospholipase C activation, intracellular calcium mobilization, protein kinase C activation, and downstream signaling cascades including MAPK, ERK, and NF-κB pathways. Through these signaling mechanisms, AT1R regulates blood pressure, vascular tone, electrolyte balance, cardiac remodeling, inflammation, and cellular proliferation.

AT1R plays a central role in cardiovascular physiology and pathophysiology and is one of the most extensively studied GPCR drug targets. Dysregulated AT1R signaling has been implicated in hypertension, heart failure, atherosclerosis, chronic kidney disease, pulmonary hypertension, fibrosis, and metabolic disorders. Consequently, AT1R antagonists, commonly known as angiotensin receptor blockers (ARBs), have become widely used therapeutics for the treatment of cardiovascular and renal diseases. Because of its clinical importance and well-characterized signaling pathways, AT1R remains a key target for drug discovery and receptor pharmacology research.

eEnzyme's Human AT1R Stable Cell Line provides a reliable and reproducible cellular platform for investigating AT1R-mediated signaling and evaluating receptor-targeted therapeutics. The cell line enables quantitative assessment of agonists, antagonists, inverse agonists, and allosteric modulators through calcium mobilization assays, reporter gene assays, and other GPCR functional screening platforms. Researchers can utilize this stable cell line for receptor characterization, compound potency determination, lead optimization, and high-throughput screening applications.

Applications of the AT1R Stable Cell Line

• Angiotensin II receptor pharmacology studies
• AT1R agonist and antagonist screening
• Angiotensin receptor blocker (ARB) evaluation
• Calcium mobilization and GPCR signaling assays
• Drug discovery and lead optimization programs
• Cardiovascular and renal disease research
• High-throughput screening applications
• Functional characterization of AT1R modulators

Biological Significance of AT1R

AT1R is the primary mediator of the biological effects of angiotensin II and serves as a key regulator of cardiovascular and renal function. Activation of AT1R promotes vasoconstriction, aldosterone secretion, sodium retention, cellular proliferation, inflammatory responses, and tissue remodeling. While these responses are essential for maintaining blood pressure and fluid homeostasis under physiological conditions, chronic AT1R activation contributes to the development of hypertension, cardiovascular disease, renal injury, and fibrosis. Because of its central role in the renin-angiotensin system and its proven clinical relevance as a therapeutic target, AT1R remains one of the most important GPCRs in cardiovascular pharmacology, translational medicine, and drug discovery research.
