The E8L protein is a highly conserved surface membrane protein of Monkeypox virus (MPXV) and other orthopoxviruses. It is the homolog of the vaccinia virus D8L protein and is present on the intracellular mature virion (IMV), where it mediates viral attachment to host cells through interactions with cell-surface glycosaminoglycans. Because E8L plays a critical role in the early stages of viral infection and is exposed on the virion surface, it has become an important target for vaccine development, neutralizing antibody discovery, and diagnostic assay development.
