Description
|
This cell line is a HEK-293-CNG cell line that expresses a recombinant human DRD1. HEK-293-CNG cells express a modified CNG (Cyclic Nucleotide Gated) channel that opens in response to elevated intracellular cAMP levels and consequently result in ion flux and cell membrane depolarization. The assay allows both end-point and kinetic measurement of intracellular cAMP changes with a FLIPR or a fluorescence microplate reader.

About Human Dopamine Receptor D1 (DRD1/D1R) Stable Cell Line
Dopamine Receptor D1 (DRD1), also known as D1 receptor (D1R), is a Class A G protein-coupled receptor (GPCR) and the most abundant member of the D1-like dopamine receptor family, which also includes DRD5. Activated by the endogenous neurotransmitter dopamine, DRD1 primarily couples to Gs proteins, stimulating adenylate cyclase activity and increasing intracellular cAMP levels. The receptor is highly expressed in the striatum, nucleus accumbens, cerebral cortex, and other regions of the central nervous system, where it regulates motor function, cognition, learning, memory, and reward signaling.
DRD1 plays essential roles in dopaminergic neurotransmission and has been implicated in Parkinson's disease, schizophrenia, drug addiction, attention deficit hyperactivity disorder (ADHD), and cognitive disorders. Consequently, DRD1 has become an important therapeutic target for neurological and neuropsychiatric drug discovery.

Biological Significance of DRD1
DRD1 is a key mediator of excitatory dopamine signaling in the central nervous system. Activation of the receptor increases intracellular cAMP through Gs-mediated signaling, regulating neuronal excitability, synaptic plasticity, motor control, cognition, and reward-related behaviors.
