G-protein coupled receptor 119 (GPR119) ACTOneTM Stable Cell Line
|
|
Description
|
This cell line is a HEK-293 CNG cell line that expresses a recombinant human GPR119. HEK-293 CNG-Slcla3 cells express a modified CNG (Cyclic Nucleotide Gated) channel that opens in response to elevated intracellular cAMP levels and consequently result in ion flux and cell membrane depolarization. The assay allows both end-point and kinetic measurement of intracellular cAMP changes with a FLIPR or a fluorescence microplate reader.

cAMP dependent human GPR119 cell-based assay;
cell-based high-throughput screening of human GPR119 agonists/antagonists.

Synonym: G Protein-Coupled Receptor 119, GPCR2, Glucose-Dependent Insulinotropic Receptor, HGPCR2, G-Protein Coupled Receptor 119, G-Protein Coupled Receptor 2
About GPR119 and Applications of the ACTOne GPR119 Stable Cell Line
G Protein-Coupled Receptor 119 (GPR119) is a Class A G protein-coupled receptor predominantly expressed in pancreatic β-cells and intestinal enteroendocrine cells. Activation of GPR119 stimulates intracellular cAMP production through Gs-mediated signaling pathways, promoting glucose-dependent insulin secretion and the release of incretin hormones such as glucagon-like peptide-1 (GLP-1). Because of its important role in glucose homeostasis and metabolic regulation, GPR119 has emerged as a promising therapeutic target for type 2 diabetes, obesity, and other metabolic disorders.
eEnzyme's ACTOne Human GPR119 Stable Cell Line provides a reliable and reproducible cellular platform for GPR119-targeted drug discovery, receptor pharmacology, and GPCR screening applications. This assay-ready cell line supports the evaluation of agonists, antagonists, and allosteric modulators, enabling receptor characterization, potency determination, lead optimization, and high-throughput compound screening. Researchers can utilize this cell line to investigate GPR119-mediated signaling mechanisms, identify novel metabolic disease therapeutics, and accelerate GPCR-focused drug development programs.
The ACTOne GPR119 Stable Cell Line is compatible with membrane potential assays and other GPCR screening platforms. Its stable receptor expression, robust assay performance, and suitability for high-throughput workflows make it a valuable tool for studying receptor pharmacology, metabolic signaling pathways, and next-generation therapies targeting diabetes and obesity.
Advantages of eEnzyme's ACTOne GPR119 Stable Cell Line
• Stable recombinant human GPR119 expression • Optimized for ACTOne membrane potential assays • Suitable for agonist and antagonist screening • Supports high-throughput screening workflows • Ideal for GPCR drug discovery applications • Consistent and reproducible assay performance
GPR119 Signaling Pathway
Upon agonist binding, GPR119 activates Gs proteins, leading to stimulation of adenylate cyclase and increased intracellular cAMP levels. Elevated cAMP promotes glucose-dependent insulin secretion from pancreatic β-cells and stimulates incretin hormone release from intestinal L-cells, including GLP-1. These biological effects have made GPR119 an attractive target for metabolic disease research and therapeutic development.
