Description
|
This cell line is a HEK-293 CNG-Slcla3 cell line that expresses a recombinant human GRM8. HEK-293 CNG-Slcla3 cells express a modified CNG (Cyclic Nucleotide Gated) channel that opens in response to elevated intracellular cAMP levels and consequently result in ion flux and cell membrane depolarization. The assay allows both end-point and kinetic measurement of intracellular cAMP changes with a FLIPR or a fluorescence microplate reader.

About Human Metabotropic Glutamate Receptor 8 (GRM8/mGluR8) Stable Cell Line
Metabotropic Glutamate Receptor 8 (GRM8), also known as mGluR8, is a Class C G protein-coupled receptor (GPCR) belonging to the Group III metabotropic glutamate receptor family. Activated by the neurotransmitter L-glutamate, GRM8 primarily couples to Gi/o proteins, leading to inhibition of adenylate cyclase, reduced intracellular cAMP production, and suppression of neurotransmitter release. The receptor is widely expressed in the cerebral cortex, hippocampus, cerebellum, thalamus, and other regions of the central nervous system, where it regulates synaptic transmission and neuronal excitability.
GRM8 functions primarily as a presynaptic inhibitory receptor that helps maintain excitatory-inhibitory balance and protects neurons from excessive glutamatergic signaling. Dysregulation of GRM8 has been associated with anxiety, depression, epilepsy, chronic pain, and neurodevelopmental disorders, making it an emerging target for CNS drug discovery.
