About Hepatitis B Virus X (HBx) Recombinant Protein

The Hepatitis B virus X protein (HBx) is a small, multifunctional regulatory protein encoded by the X open reading frame (ORF) of the hepatitis B virus (HBV) genome. Although HBx is not a structural component of the virion, it is essential for efficient HBV replication and productive infection. HBx functions primarily by interacting with numerous host cellular proteins to regulate viral transcription, signal transduction, protein degradation, DNA repair, apoptosis, autophagy, and cell cycle progression. One of its best-characterized functions is the recruitment of the DDB1-containing E3 ubiquitin ligase complex to promote degradation of the host restriction factor Smc5/6, thereby relieving suppression of HBV transcription from covalently closed circular DNA (cccDNA).

eEnzyme's Hepatitis B Virus X (HBx) Recombinant Protein is expressed in HEK293 cells, providing native mammalian folding and post-translational modifications that closely resemble the natural viral protein. The purified recombinant HBx is suitable for ELISA, Western blotting, antibody characterization, protein-protein interaction studies, drug screening, HBV replication research, and host-virus interaction studies. Its mammalian expression preserves conformational integrity, making it an excellent reagent for investigating HBx-mediated cellular signaling and viral pathogenesis.

•  Protein-protein interaction assays
•  ELISA and Western blot analysis
•  Antibody characterization
•  Drug discovery and inhibitor screening
•  HBV pathogenesis research
•  Hepatocellular carcinoma (HCC) studies

Biological Significance of HBx

HBx is a key regulatory protein that is indispensable for productive HBV infection. Rather than acting as a DNA-binding transcription factor, HBx modulates host cellular pathways through interactions with numerous cellular proteins. It activates multiple signaling pathways—including MAPK, NF-κB, PI3K/Akt, and JAK/STAT—and regulates transcription, apoptosis, autophagy, mitochondrial function, and DNA damage responses. HBx also counteracts the host restriction factor Smc5/6 by recruiting the DDB1-CUL4 E3 ubiquitin ligase complex, thereby promoting Smc5/6 degradation and enabling transcription from HBV cccDNA, a critical step in the viral life cycle.

Persistent expression of HBx has also been implicated in the development of hepatocellular carcinoma (HCC) through its effects on genomic stability, chronic inflammation, epigenetic regulation, and cellular proliferation. Consequently, HBx remains a major target for studies of HBV biology, chronic hepatitis B, liver cancer, and the development of next-generation antiviral therapeutics. The Hepatitis B Virus X (HBx) Recombinant Protein provides a high-quality research reagent for investigating HBx function, identifying host interaction partners, evaluating therapeutic inhibitors, and advancing HBV virology, immunology, and translational liver disease research.
