Human Papillomavirus Type 16 (HPV16) is a high-risk papillomavirus strongly associated with cervical cancer, anogenital cancers, and a subset of head and neck cancers. The E6 protein is one of the major HPV16 oncoproteins and plays a critical role in viral-mediated carcinogenesis. E6 promotes the degradation of the tumor suppressor p53 through interaction with the cellular ubiquitin ligase E6AP (UBE3A), resulting in inhibition of apoptosis, genomic instability, and uncontrolled cell proliferation. Together with the E7 oncoprotein, E6 is essential for the initiation and maintenance of HPV-induced cellular transformation.
