Description

This cell line is a HEK-293-CNG cell line that expresses a recombinant human MC5R. HEK-293-CNG cells express a modified CNG (Cyclic Nucleotide Gated) channel that opens in response to elevated intracellular cAMP levels and consequently result in ion flux and cell membrane depolarization. The assay allows both end-point and kinetic measurement of intracellular cAMP changes with a FLIPR or a fluorescence microplate reader.

About Human Melanocortin 5 Receptor (MC5R) Stable Cell Line

Melanocortin 5 Receptor (MC5R) is a Class A G protein-coupled receptor (GPCR) that is activated by endogenous melanocortin peptides, including α-melanocyte-stimulating hormone (α-MSH), β-MSH, γ-MSH, and adrenocorticotropic hormone (ACTH), which are derived from the proopiomelanocortin (POMC) precursor. MC5R primarily couples to Gs proteins, stimulating adenylate cyclase activity and increasing intracellular cAMP levels. Unlike other melanocortin receptors, MC5R is widely expressed in both the central nervous system and peripheral tissues, including sebaceous glands, exocrine glands, skeletal muscle, adipose tissue, immune cells, and skin, where it regulates exocrine secretion, energy metabolism, lipid homeostasis, and immune function.

MC5R plays important roles in sebaceous gland function, thermoregulation, lipid metabolism, skeletal muscle physiology, and inflammatory responses. Dysregulation of MC5R signaling has been associated with metabolic disorders, inflammatory diseases, skin disorders, and impaired exocrine gland function, making the receptor an emerging therapeutic target for metabolic and inflammatory disease research.
