About Phlebovirus Glycoprotein Gc

Phleboviruses are enveloped RNA viruses belonging to the family Phenuiviridae and include important human and veterinary pathogens such as Rift Valley fever virus (RVFV), Severe Fever with Thrombocytopenia Syndrome Virus (SFTSV), and Heartland virus. Viral infection is mediated by two envelope glycoproteins, Gn and Gc, which function together to facilitate host-cell attachment, viral entry, membrane fusion, and virion assembly.

Glycoprotein Gc is the principal viral fusion protein responsible for mediating fusion between the viral envelope and host-cell membranes during infection. Following receptor-mediated endocytosis and exposure to the acidic environment of endosomes, Gc undergoes extensive conformational changes that drive membrane fusion and enable release of the viral genome into the host cell cytoplasm. Because of its essential role in viral entry, Gc has become a major focus of structural biology, antiviral drug discovery, vaccine development, and neutralizing antibody research.

Biological Significance of Glycoprotein Gc
