About Human Enterovirus A71 (EV71) VP4 Protein

Human Enterovirus A71 (EV71) is a major causative agent of hand, foot and mouth disease (HFMD) and is associated with severe neurological complications including meningitis, encephalitis, and acute flaccid paralysis. The VP4 protein is one of the four structural proteins that comprise the EV71 capsid and is generated during virion maturation through cleavage of the precursor protein VP0. Unlike VP1, VP2, and VP3, which are exposed on the viral surface, VP4 is located on the inner surface of the capsid where it contributes to capsid stability, virion maturation, and the early stages of infection.

VP4 plays a critical role during viral entry. Following receptor engagement, the EV71 capsid undergoes conformational changes that result in the externalization of VP4 and the N-terminus of VP1. Together, these proteins help create a membrane pore that enables transfer of the viral RNA genome into the host cell cytoplasm.
