Ebola Virus

Ebola disease is a severe viral illness caused by members of the genus Orthoebolavirus in the family Filoviridae. Several orthoebolaviruses can cause disease in humans, including Ebola virus, Sudan virus, Bundibugyo virus, and Taï Forest virus. These enveloped viruses are important targets for infectious disease surveillance, vaccine research, therapeutic antibody development, and antiviral drug discovery.

The viral glycoprotein (GP) forms trimeric spikes on the virion surface and mediates host-cell attachment, endosomal entry, and membrane fusion. Because GP is the principal antigen exposed on the viral surface, it is the major target of neutralizing antibodies, vaccines, and entry-inhibiting therapeutics. The nucleoprotein (NP) encapsidates the viral RNA genome and is widely studied in viral replication, diagnostics, and host immune-response research.

Ebola Virus Genome and Encoded Proteins

The Ebola virus genome is a non-segmented, negative-sense, single-stranded RNA molecule approximately 19 kb in length. It contains seven major genes encoding the nucleoprotein NP, polymerase cofactor VP35, matrix protein VP40, glycoprotein GP, transcription activator VP30, minor matrix protein VP24, and RNA-dependent RNA polymerase L. Coordinated interactions among these proteins control genome replication, transcription, virion assembly, budding, immune evasion, and viral entry.

Recent Advances in Ebola Virus Research

Major progress has been achieved in the prevention and treatment of disease caused by Zaire ebolavirus. The rVSV-based ERVEBO® vaccine is licensed for prevention of disease caused by Ebola virus, and the monoclonal antibody therapies Inmazeb® and Ebanga® are approved treatments for Zaire ebolavirus infection. Long-term studies have also demonstrated durable vaccine-induced cellular and humoral immune responses, supporting continued investigation of booster timing and immune correlates of protection.

Current research increasingly focuses on broad-spectrum vaccines and antibody combinations capable of protecting against multiple orthoebolavirus species. This remains especially important because licensed vaccines and treatments are not yet available for all human-pathogenic orthoebolaviruses, including Bundibugyo and Sudan viruses. Structure-guided antigen design, conserved GP epitope targeting, mRNA vaccine platforms, and cross-reactive monoclonal antibodies remain active areas of research.
