About Human Metapneumovirus
Human metapneumovirus (HMPV or hMPV) is an enveloped, non-segmented, negative-sense single-stranded RNA virus in the genus Metapneumovirus and family Pneumoviridae. It is a common cause of upper and lower respiratory tract illness in people of all ages, with the greatest risk of severe disease among young children, older adults, and individuals with weakened immune systems.
HMPV virions are pleomorphic and surrounded by a host-derived lipid envelope containing three membrane-associated proteins: fusion protein F, attachment glycoprotein G, and small hydrophobic protein SH. F mediates fusion of the viral envelope with the host-cell membrane and is the principal target of neutralizing antibodies. G contributes to attachment, while SH is associated with virus-host interactions and modulation of cellular responses.
Beneath the envelope, matrix protein M supports virion organization and assembly. The genomic RNA is encapsidated by nucleoprotein N to form a helical ribonucleoprotein complex associated with phosphoprotein P, the large RNA-dependent RNA polymerase L, and the M2-1 and M2-2 regulatory proteins.

Genome Organization & Encoded Proteins
The HMPV genome is approximately 13.3 kb and contains eight genes arranged in the order 3′-N-P-M-F-M2-SH-G-L-5′. These genes encode nine proteins because the M2 gene contains two overlapping open reading frames that produce M2-1 and M2-2. Unlike members of the genus Orthopneumovirus, HMPV lacks the NS1 and NS2 genes.
