Description

This cell line is a HEK-293-CNG cell line that expresses a recombinant human CHRM4. HEK-293-CNG cells express a modified CNG (Cyclic Nucleotide Gated) channel that opens in response to elevated intracellular cAMP levels and consequently result in ion flux and cell membrane depolarization.

About Human Muscarinic Acetylcholine Receptor M4 (CHRM4/M4R) Stable Cell Line

Muscarinic Acetylcholine Receptor M4 (CHRM4), also known as the M4 muscarinic receptor (M4R), is a Class A G protein-coupled receptor (GPCR) that is activated by the endogenous neurotransmitter acetylcholine. CHRM4 primarily couples to Gi/o proteins, resulting in inhibition of adenylate cyclase activity, reduced intracellular cAMP production, and modulation of ion channel activity. The receptor is highly expressed in the striatum, cerebral cortex, hippocampus, and other regions of the central nervous system, where it regulates cholinergic and dopaminergic neurotransmission, motor function, cognition, and synaptic plasticity.

CHRM4 has emerged as an important therapeutic target for schizophrenia, Parkinson's disease, substance use disorders, Alzheimer's disease, and other neurological disorders. Selective M4 agonists and positive allosteric modulators (PAMs) have attracted significant interest because they may provide antipsychotic efficacy while reducing the adverse effects associated with conventional dopamine receptor antagonists.

Biological Significance of CHRM4

CHRM4 is a key regulator of cholinergic signaling in the central nervous system. Activation of the receptor inhibits intracellular cAMP through Gi/o-mediated signaling and modulates the release of acetylcholine and dopamine, particularly within the striatum.
