About the Elite™ Potassium Ion Channel Assay Kit
Potassium (K⁺) channels are a diverse family of membrane proteins that regulate cellular excitability, membrane potential, neurotransmitter release, hormone secretion, muscle contraction, electrolyte balance, and cell proliferation. Because potassium channels play critical roles in cardiovascular, neurological, metabolic, and immune system function, they have become important therapeutic targets for the treatment of arrhythmias, epilepsy, pain, diabetes, dementia, and other diseases. Quantitative measurement of potassium channel activity is therefore widely used in ion channel research, drug discovery, and high-throughput screening applications.
eEnzyme's Elite™ Potassium Ion Channel Assay Kit provides a no-wash, homogeneous, cell-based assay for high-throughput measurement of potassium channel and transporter activity. The assay utilizes thallium influx as a surrogate indicator of potassium ion channel activity and employs a proprietary fluorescent dye with high affinity for thallium ions. Upon channel activation, thallium enters the cell through open potassium channels and generates a robust fluorescent signal proportional to channel activity. This sensitive fluorescence-based approach enables rapid and quantitative analysis of potassium channel function without the need for radioactive tracers or electrophysiological measurements.
The Elite™ Potassium Ion Channel Assay Kit is optimized for 96-well and 384-well microplate formats and can be readily adapted to automated high-throughput screening workflows. The assay is compatible with FlexStation® instruments and standard fluorescence microplate readers, providing a convenient platform for screening channel modulators, evaluating compound potency, characterizing ion channel pharmacology, and investigating potassium transporter function. Its homogeneous assay format minimizes handling steps and assay variability while supporting efficient large-scale screening programs.
Applications of the Potassium Ion Channel Assay Kit
• Potassium channel activity measurement • Potassium transporter functional assays • Ion channel pharmacology studies • Agonist and antagonist screening • Drug discovery and high-throughput screening • Compound potency and efficacy determination • Electrophysiology alternative screening assays • Neuroscience, cardiovascular, and metabolic disease research
Biological Significance of Potassium Channels
Potassium channels are essential regulators of membrane potential and cellular excitability. By controlling potassium ion flux across the plasma membrane, these channels influence action potential generation, cardiac rhythm, neuronal signaling, hormone secretion, smooth muscle contraction, and many other physiological processes. Dysfunction of potassium channels has been implicated in numerous disorders including cardiac arrhythmias, epilepsy, chronic pain, hypertension, diabetes, and neurodegenerative diseases. Because potassium channels represent one of the largest classes of ion channel drug targets, reliable functional assays are critical for understanding channel biology, identifying therapeutic compounds, and evaluating potential off-target effects during drug development. Fluorescence-based potassium channel assays have become valuable tools for ion channel screening because they provide sensitive, quantitative, and high-throughput measurements of channel activity in living cells.
