SARS-CoV-1, MERS-CoV and SARS-CoV-2 belong the subfamily of Coronaviruses. SARS-CoV-2 is a novel human pathogen that spread into a pandemic COVID19 that has caused large number of deaths worldwide and immense economic damage since the start of the outbreak in late 2019.
Other human pathogenic coronaviruses known include HCoV-NL63, HCoV-229E, HCoV-OC43 and HKU1, but SARS-CoV-1, MERS-CoV and particularly SARS-CoV-2 have been the most worrisome. They usually attack the respiratory, intestinal, and maybe nerve systems of their hosts.
Coronaviruses contain a positive-sense, single-stranded RNA genome. The genome size ranges from 26.4 to 31.7 kilobases and is among the largest RNA viruses. The genome is polycistronic with a large pre-polypetide translated that is processed by proteolytic cleavage to most of the viral structural and proteolytic processing proteins. Of particular interest is the Spike (S) protein that mediates cell entry and gives the virus its typical “corona” around the viral envelope in which it is embedded when visualized under EM.

The ORFs 1a and 1b encode the replicase polyprotein (pp1ab). The replicase polyprotein self cleaves to form 16 nonstructural proteins (nsp1–nsp16). The later ORFs encode the four major structural proteins: spike, envelope, membrane, and nucleocapsid. Interspersed between these ORFs are the ORFs for the accessory proteins. The number of accessory proteins and their function is unique depending on the specific coronavirus.
SARS-CoV-2 is a 125nm-diameter, spherical-shaped β-coronavirus. It has four structural proteins, known as the S (spike), E (envelope), M (membrane), and N (nucleocapsid) proteins; the N protein holds the RNA genome, and the S, E, and M proteins together create the viral envelope.
